The idea with speedrunning is that you want to go as fast as possible right? Attaching rules like no glitches or only ending C kinda defeats the purpose of that. Some people will argue that doing anything but the fastest category or 100% is pointless
I've ran a game with different difficulties on hard for three years now because other people ran co-op of the game on hard and so I felt like I'd do that too, and while I don't really regret doing so, if I could start over I'd most likely run it on the easiest difficulty from the start. On harder difficulties you'll probably find a lot lot more so called run killers than you would on any easier difficulty which is why I tend to reset every run where I die on the first three missions.
I don't think anyone hates me for doing it though so yeah it's up to you. Run whatever category on whatever difficulty you want anon, best of luck
