well, there are a couple of parts where it's pointed out that there is homoerotic context floating around (i don't want to spoil it for you, but at some point other people freely interpret Sam and Dean as an item).
In the first season in one of the first episodes they are mistaken as a gay couple, which is more comical than anything.
There is also that Castiel is so willing to go out of his way for Dean, and I guess that could be seen as something more.
So I guess a few ambiguously interpretations can be made.
