Well it's never been officially stated to be canon. Also, I don't remember this being said in the manga but I might have just forgotten, I noticed that Tonio said he doesn't have any siblings in the anime which is contradictory to PHF. Either way, I like to think it's canon. Araki helped with it and it's a really good addition to the story and a perfect ending for Vento Aureo. Hopefully it'll get an OVA when Vento Aureo gets animated.

The entire narrative revolves around it. It's literally called "The Killing Joke". The joke that finally brings batman to the realization that he has to kill the Joker because there is no chance of rehabilitation for him. Afterwards him and Carrol roamed around trying to help people. They ended his comic, and threw him in the Omega Men mix.
