IIRC you need to use some kernel patch or something for the card to start. It is on the page that's on the front of the card.
After that is just dumping roms at the root of the SD and booting em up.

When I still used those cards I remember using lolSnes for SNES emulation and Neo DS for some Neo Geo games.
Never managed to run GBA since those needed a Dstwo card that uses the second slot.

It does. However Desmume has the option for hardware acceleration. That mean that if you've got a nice rig you can run games at much higher settings. Also NO$GBA kinda glitched with my mouse when I used it
