Manus definitely isn't harder. I can't comment on Flamelurker because DeS was my first Souls, so it's hard for me to judge from my own experience. I killed Flamelurker first try every run after my first.

Had a harder time with the pthumerian descendant and abhorrent beast in chalice dungeons.
In the DLC specifically ludwig and even maria for some reason gave me more trouble than laurence and kos. This was my run with a blood level 80 bloodtinge character.
My friend who runs a str build had no trouble with any of these bosses cause he just stagger locks everything into easy mode

Flamelurker is a great boss but he's small time. I recently played through DeS again and 1-shotted him easily. I'm not sure why I had as much trouble as I did before. Probably because DeS was my first souls game I got back when it first came out. False King is definitely harder than Flamelurker though.
