Well in the comics Jameson is revealed to secretly be jealous of Spider-Man, this was a nod to that. Shame about how they fucked up the scene though.

It was a reference to Jameson's Last Scoop, where he finally kills Spider-Man, unmasks him, realizes who it was and assumes the identity to make up for the death of the poor kid.

Raimi wanted it to be a joke in the movie, but it turned out JK Simmons actually looked good in the suit. It would have been funnier if JK was fat or something.
