I'm not sure how Spidey's spider-sense would work with super speed. I've read multiple comics where the opponent has super speed, and it flip-flops between being irrelevant since his spider-sense can detect literally ANY movement, or being unable to pick up the ultra fast movement.

You also have to account for how much the heroes know about each other, how much time they have to prepare, their intent or motive. The who, what where, when, why and how are largely ignored which just leads to unoriginal and boring discussions. I am going to propose some criteria that will hopefully make this conversation more lively and if not it was at least fun to write and think about.
Criteria 1: Location
I dislike the arena style matchup because i feel that it takes away strengths from certain heroes and unjustly inflates the others. All fights will be one on one. A single superhero will have identical clones with the same knowledge as the original, they will have no knowledge of each other. Each clone will go to/invade a single city/area where an opponent resides.
i.e. Batman A will fight cap in his city, Batman B will fight hulk in his city etc. In the above scenarios involving batman. Batman is the invader while cap and hulk are defenders. Do this for all possible combinations and tally up the winners. After these fights occur the playing field will be reset. Their can be ties. The tie situation is mentioned under Criteria 2. Neither side knows that their universe will reset after the fight is finished.

Criteria 2: Knowledge
The invading character will function as a villain. Their intent will be to kill or incapacitate the other hero, or remove them from their city for more than 1 month straight. If both heroes are killed or incapacitated then this will result in a tie. The characters keep their original moral attributes. Basically what personality they started off with. The reason they decided to be superheroes. If a character was ok with killing before he/she became a superhero then they will retain that trait for the remainder of the duel. If a hero is ok wih taking hostages then he shall be able. Anyways i kinda sidetracked a bit. The invading character will function as a villain. The defending hero will not have knowledge of the invader until the fight occurs. The invader cannot recruit help from other heroes or villains (unless this is what he commonly does). If a hero usually uses a source in the police department then that source shall be available. The invading character will be given shelter at an undisclosed building and given his/her regular wardrobe along with relevant I.D's/Credentials. This initial setup will not alert the defending hero. Actions that the invading hero takes may put him in the radar of the defending hero. If a hero commits an act that raises his attention then the chances of him being discovored also increases. While the invader is inside the city he can uses whatever resources he can scour to gather intel on his opponent. If the defender is attacked and somehow the fight does not immediately end, depending on his intelligence he can find out information about the invader.

Criteria 3: Timeline and Universe
This is probably the hardest part for me to write and might be confusing. If you need confirmation on something let me know and I will try to explain as best as I can.
Each hero has an origin. They have motivations and fears. A majority of them have a moment that broke them, a moment that pushed them over the edge to become heroes. The invading hero and the defending hero will come from the original earth. Before the multiverses were mentioned. The heroes that will be chosen from this battle have not faced any world endig dillemas (this usually leads to some pretty disgusting power creep). All of them will have discovered their powers but its not really refined. They know what they can and can't do but there are some nuances that they can't get a hold of. This should take care of any over powered heroes, but it won't hurt them too badly.
There are some writers that are a bit...creative and overzealous with their portrayal of these heroes. There will be no plot armour in this battle. The more comvoluted and complicated the plan the more likely it is to fail. There are times where some 'weaker' heroes could win. but thats not really the point. We should care about the averages. Just batman can kill cap by sliding small pieces of radioactive uranium up his ass for the next 12 years doesn't mean that he will win. The chances of this happening without notice are highly unlikely so be a bit reasonable. Now if batman is able to consistenly outsmart cap in fights then he would take the win.

Nothing is infinite there are limits (looking at you flash).
The speed limit for all heroes is under light speed. Though with the criteria set above i don't think that will be reached by anybody.
Prep time cannot,should not and will not defy logic.
On average if a matchup is power vs no power. The hero with powers has a huge advantage.
Kryptonite is available at all museums. Will the heroes capitalize on this weakness? Will this weakness even matter?
