It would have to be really damn persuasive, since without the astronomican the imperium would collapse pretty much instantly.
Even putting aside the military implications (not being able to conduct fleet space warfare or move troops and ordinance around), a lot of planets are wholly reliant on cargo shipments for food that they can't produce on-planet themselves, e.g. forge worlds and hive cities. Not to mention other logistics stuff e.g. medical supplies, other tech...
And aside from all that, the imperium loses whole planets from time to time even when everything is functioning perfectly, because of the beurocracy. How could the imperium continue to exist as a beurocratic entity with no starship travel?

The Emperor's overpowered psyker abilities allows him to keep the Astronomican, a beacon / lighthouse in the warp, lit from wherever he is marking where Terra is and allowing navigators to have a point of reference in the warp. It is probably part of the reason a certain Ork could have been the end of him hadn't Horus been there.
Of note is that the Astronomican only lights the "western" part of the galaxy for 50 000 light years, the Milky Way is somewhere between 100 000 and 180 000 light years across.
It is fully possible to travel without the Astronomican but is hellishly hard for the navigator as they will lack a "easy" point of reference.
The Emperor must be on the Golden throne as he keeps the daemon infested webway portal controlled by the Golden Throne shut. If his body dies we enter speculation territory but his psychic presence keeping the Astronomican lit may scatter across the warp and there is daemon party everyone is invited to on Terra.
Malcador had to be on the throne during the final siege on Terra to keep the daemon party out.
Some margins for error but give the Lexicanum a once through.

The reason the astronomicon fails when the emps dies isn't that he isn't there to make it but rather that the human webway entrence will be unsealed and ALL the deamons spill out on to terra talking it as a demonworld and destroying the primary choir in the process.
Tldr you can have the astronomicon without the emps but not terra coz that's where we keep all the psykerz and there stuff.
