Immediately call in all reserves from the countryside, and recruit older men and teenage boys to fight/support. Do the same with women that want to help make uniforms, tend the wounded, etc. Equip all with spears, shortswords, and crossbows.
Try to hire mercenaries by taking out loans. The defenses would need all the crossbowmen it could get to man the walls.
Fortify the city's defenses as best as I could. Scrap wood and stone in the city to create catapults and trebuchets to counter the siege engines the Turks will have.
Request aid from the pope and the Catholic powers, it may not do much, but asking never hurt. Food alone would help.
Outfit a ragtag navy with those dope greek fire cannons and try to pin their navy in the water outside the city.
