It could only happen in a state where a huge majority supports secession. Enough so that the whole apparatus of the state government, politicians down to law enforcement are on board.
In that case the federal government would have a PR blitz to try and convince them that it would be catastrophic if they leave. Threaten them with every manner of economic pain if they leave. If they insist on doing it anyway and then take concrete steps (like taking over US military facilities and federal offices) then it'll provoke a violent response most likely. You never know though. If we have an Obama or Bernie Sanders in the WH they might let it happen. But no president wants to be known as the guy who broke the Union. You would also start getting other secession movements springing up if one appears to be successful. It can't be allowed to happen.
