Because a lot of good comedy comes from pain and self-deprecation. It comes from being an utter scumbag and putting all of that out there in a humorous way.
Women cannot do this. Women crave acceptance. They want everyone to think they're great. They are also terrible storytellers compared to men so they rely on gimmicks (usually "I'M A HUGE SLUT TEE HEE") and generally come off as incredibly fake, laying on the shtick far too thick.

I think a big part of it all is the fact that comedy is subjective. Now saying that, there still are plenty of shit comedians of either sex... however the best comedians always draw from observations about society and how different people interact. The problem is women often lead a very passive lifestyle. They lack the same sort of ambition and are coddled a bit more. That's why the few women that ARE funny *cough* Maria Bamford *cough* have history of mental illness, familial abuse, or other shit. Another reason why "muh vagina" jokes aren't funny and thus a lot of female comedians aren't, is the fact that it's harder for people to "get with" the jokes, especially men. We can't empathize with that, meanwhile male comedians when they talk make jokes about male anatomy do so in an objective and often self ridiculing or humiliating way and thus everyone can get something out of it.
