No it is not.
The advantage to FLAC is not that it sounds better, but that if you want to recode your file to another format, you won't lose more quality, as you would with the 320 files.
It has been proven numerous times that the human ear can't hear the difference, with audio engineers and audiophile geeks on the best possible setups available. Google it.
