Given how close Times Square has already gotten to a bombing, I'd say pretty serious. How hard is it to drive a semi truck through Times Square? Anyone can do it. The driver himself doesn't even need to be aware of the bomb.

That's pretty much it. But it's managed to become an icon of NYC, a place where people tend to congregate to shop and eat. It's also the only interesting thing in midtown (compared to lower Manhattan).
For a terrorist it's an amazing opportunity as there's always a largeish crowd of people standing around, while regular car/truck traffic can rush by. Which means a full size truck bomb (50 foot intermodal container) can be set off and inflict a massive amount of causalities.

Regulating truck traffic is (at least in my experience) the domain of the Port Authority as they're the ones with the majority of it (due to being a port).
