I got a Samsung Galaxy Note 4, and I'm with android 5.0.1 root.

How can I do the update to 6.0.1 without problems ?
Still root would be perfect also.

You can't. You Have to backup and restore your phone.

Restoring my phone will allow me to update to 6.0.1 unroot ?

Make a back up.

Completely wipe everything.

Phone will not have root on it so it'll be fine to update. Previous root will not affect it. Unless you didn't wipe your phone properly
