It's kind of a double edged sword:

For hardware reasons, some people say you are prematurely wearing out the internal parts(especially spinning hard drives) or letting them wear down due to heat buildup if you leave it on all the time. The other school of thought is that the turning on and off of a machine causes power surges, and each component only has so many power cycles before it pops.

Software-wise, there are a lot of advantages to turning it off. Some programs develop "memory leaks" where they set aside a block of memory every time they start, and sometimes forget to release it when they're done. Rebooting your machine clears out memory leaks. Also, if you have automatic updating set on your PC, many updates are not fully applied until after a reboot. New updates are prevented from being loaded until the last one has completed being installed (machine rebooted). There is also the vulnerability issue that Wounded Duck stated, but if you have a good firewall in place and the proper antivirus software, you should be OK.

Lastly, there is the electricity bill: A PC with a 300 watt power supply and a monitor hooked up, 24x7 will suck a lot of juice out of a wall. It's like having a 300 watt bulb on all the time, plus the monitor can be like another 60 watt bulb if it's a CRT monitor.

If you don't care about the electricity bill, and you average a new computer every 3-4 years, my suggestion is to do what most IT departments do with non-essential servers: Leave it on, but make a practice of rebooting it once a week to clear out potential memory leaks and allow updates.
