OP isn't talking about merely tunnelling Bittorrent or something over Tor, but about some kind of P2P app specifically designed with mixnets?
Tor itself isn't a great choice for high-bandwidth node/node connections because of the routing algorithm. That will partially change with the new onion service ed25519 design, because the circuits are shorter (not basically two whole circuits joined end-to-end, like now).
I2P, however, actually is designed for node-to-node connections - proof of concept, Syndie - and has variable delay.
It's not precisely what I would do if I were to redesign it today, because of the global passive attacker problem of all plain onion routing and plain garlic routing, but I can't say that my current designs in research are all that far away.
