When the Saint of Killers gets the nuke dropped on him, he says "Not Enough Gun" so that means a gun probably CAN hurt him.
We also know that he is affected by The Voice, provided he hears it.
The real problem is the guns. He can draw them faster than the human eye can track and he never misses. Cassidy appears to be immune, but that's because he's already dead, not to mention Ennis said he'd change it.
So, if you could somehow say "Drop the guns" in The Voice before he shot you, then shoot him, you might be able to kill him. It might be the only way to beat him is a duel.
