The radio show was made into a series of MGM film shorts, including one that debunked myths and old wives tales. The line was from that.
MGM, you'll remember, is the same production company that made Tom and Jerry, and they had the audio. Audiences who watched the Tom and Jerry shorts in the original theatrical run would have been very familiar with it, and probably thought the reference was funny.
Kind of how like when Bugs Bunny said "What's up, Doc?" for the very first time, theater audiences broke out in riotous laughter. It was a joke that they got, not a catchphrase.
