You cannot control what someone else thinks.
Generally speaking, unless you make some big deal out of asking or a big deal out of being rejected, it won't affect anything. This is not true for all people.

Test the waters without getting in too deep.
Have a non-date date. The next time you're chatting say something like "It's been fun but we have to get back to work. Would you like to get together for coffee/coke/drink after work to continue the conversation?"
If that goes well, you can ask her out with more confidence.
