The minimal requirement is 3+ other characters interested in the MC, but that's generally overly broad as there are examples that meet that criterion but are pretty certainly not harems.

If the MC predominately interact with females compared to males then it's a harem. It doesn't need the females show romantic interest toward MC. That's up to doujin and fan fiction writers.
