It could work under those circumstances. But not in the available source excluding the shitty PSP game. Taiga/Ryuuji are OTP for all that material.
But there are obvious flags which could've been taken if things had been different.
Like when she asks him to walk with her on the beach for example, instead he choose to chat with the psycho.
Could've easily been a strong flag for Ami's route.
If Ami was the main girl and it was her route, she would've been best for the Ryuuji of her own route (who would've been different as well).

the way I see it, Ryuuji 'fell' for Minori in the first place because she resembled his mother so much, a woman he admired to the core despite everything. He later fell for Taiga because of everything they'd been through and because their personalities worked so well together.
Ami is almost just there to be the audiences spokeswoman, she takes a step back and sees everything for what it is and sees it all going to shit but even if all 3 had equal chance I don't think they would have worked together, but that's more of a gut feeling than anything concrete.
