Just tried that (there are two microphones actually) and it really mutes the sound almost completely. But you have to do it properly, otherwise it's useless.
I think it would be great if Apple added a hardware LED (meaning it can't be turned off by software, not even theoretically) to both their camera and microphone. I believe with the cam this is already the case, but it would be a good move to specifically stress that, as I see a lot of people being paranoid about that.
