So, the Grand Classes (Ascension) are Servants who were a part of the original Holy Grail War (the one that Fuyuki's system is just a watered down version) and they are Heroic Spirits who are chosen and powered by the Root and are supposed to be fighting against those threats that will wipe out mankind.

No, Grand is just the strongest of servants from each class. They're not boosted, Solomon wasn't even sent by the root or the world, he just summoned himself because he's that broken.

Shouting the unreasonable, Gilgamesh fired countless noble phantasms from the Gate of Babylon opening behind him. It seemed that this rain of blades mixed with high rank noble phantasms would leave nothing remaining of Archer. But he waved the bow he was holding with his left hand and knocked aside the noble phantasms that had been fired at a speed that defied the common sense of heroic spirits.

The Heroic Spirit who deflected dozens of noble phantasms without a scratch silently held out his hand towards Gilgamesh. And then, he turned it over palm side up and curled his fingers back and forth, beckoning for Gilgamesh to come. A good way to guess, even if it's not an exact science, if it will do bonus damage is to ask yourself if the enemy you're about it use it on is an historical figure who did really exist or just a myth, if he's a myth, it will do bonus damage. But there's some quirks, for example it will do bonus damage against Vlad III because he's summoned as Dracula the vampire, who did not exist.
In the same way Enuma Elish doesn't do bonus damage against servants who changed the world with their discoveries or works of the mind, Drake, Tesla or Mozart for example won't receive any bonus damage.
