The line of succession is based on Bobby's claim of Targ blood, so technically they'd have to go to any trueborn descendants of that Targ grandma Robert had, of which there are none since Shireen, Stannis and Renly are dead, so it'd then have to go back to Targ descendants.
A Great Council would have to be called to decide which other Targ descendant, Aegon V's sisters for example had kids so descendants of that are possible.
But most likely in the show there'd be a civil war type thing where the Tyrells claim the throne and try and hold it and be supported by the people of King's Landing and Sparrow in holding onto power and not having the Lannister armies in the city slaughter them and claim it for Lannisters

If the Tyrell's or Arryns could make a claim and an alliance with each other, sure. As it stands the Tyrell's are likely to benefit from a Lannister crown though so unless they could make a play for themselves, they aren't going to back anyone but Lannister. Ditching House Lannister to back House Arryn would be a risky switch and since the Lannisters already hold KL it would make more sense to stick it out.
The Westerlands have the most battle hardened host south of the neck, the north is in civil strife, the Reach has the most men and the most resource wealth. Arryn is the freshest but the knights of the Vale wouldn't be successful alone against the two.
There isn't really a workable anti-Lannister scenario unless the Tyrell's find a way to make a claim for themselves and enlist Dorne and the Vale against House Lannister.
