This is an oversimplification of the Tinker doctrine.
While it's true that students don't surrender all of their first amendment rights at the schoolhouse gate, it is equally true that these rights are not nearly as broad and inclusive inside of school as the are outside of school. The reason for this, according to the Supreme Court, is the need for school administrators to maintain order and discipline within the school.
Thus, if they can demonstrate that this kid's speech caused or reasonably could've caused a substantial disruption, then they are well within their rights to punish him, without worrying about infringing on his first amendment rights.

This XY/XX chromosome meme needs to die. It's an extreme oversimplification and both sides of the argument are wrong about it - biological sex is tied to more things than just those chromosomes.
Same story with hormones, it's not just testosterone and estrogen, there's a whole clusterfuck of them interacting, they're just the most prominent in this case. So dumping extra of either, or simply blocking production of one, will not change someone's sex and could (will) cause a whole bunch of problems for them down the line.

Since these were Twitter posts, I should add that what federal courts are still working out is whether postings on social media made outside of school can nevertheless be punished within the school without running afoul of the first amendment.
It's all very fact-specific, but the general trend seems to be that courts will rule in favor of the school if the speech posted online "reaches" the school in some way.

I think that's a good point, and why these cases are so fact-specific.
Hypothetically, under these facts, you could image the tweet nevertheless reaching the school if it were to start a rumor that led to a fight on school grounds. If that were to happen, I think the school could punish him without worrying about the 1st A.
