Different states hold elections differently, and from those elections, based on the votes of the people (usually) delegates are selected to vote in the national Republican convention. It's the delegates who decide who wins the nomination. Contractually, most of those delegates are obligated to vote in a certain way. For example, if Trump gets the most votes in a winner-take-all state, delegates from that state will be contractually obligated to vote for Trump at the convention to try to make him the party nominee.
At the convention, if, once all the delegates cast their vote, there still isn't a candidate with more than 50% of the delegates supporting them, there is a re-vote. For the re-vote, a lot of the delegates become 'unbound' (they can vote however they want, even if that's entirely different from how their state elections went).
Most delegates are establishment people, and don't want Trump to win the nomination. They're obligated to vote for Trump in the first round of voting at the convention, but if Trump can't get more than 50% of all delegates in that first vote, they'd rather vote for someone else instead. They could give the nomination to Cruz even though fewer people voted for him, or they could completely change the rules of the RNC so that they could pick someone who didn't even run for election.
