There's been some testing but nothing extensive. So far tests have shown that 20.3 has either no penalty or might have 0/-1/-2 depending on the classes. Basically just 20.3 your CLs unless you're in combined fleet and/or sniping DDs.

Imagine if the random nodes will have BB Hime as flagship of one and Elite Ne as flagship as another. And imagine that if you sink them enough times the escort BB Hime and Elite Ne will just not be there anymore, leaving a 4 ship boss node. Or maybe they would get replaced by DDs.
I think that would be a pretty interesting use of the mechanic, although obviously it would probably be too strong if you could literally remove a BB Hime from the boss node.
