Distinct cultures will absolutely emerge, especially if communication between systems isn't immediate. Examples of this happening on a smaller scale include the Yankees of 1776 being different from the colonists of Jamestown. Given time and no outside interference, every system will have it's own culture.
It's also worth noting that local culture won't just go away entirely, it just won't be the culture of the land anymore. As I said before, a good example of this is German-majority cities in America having Oktoberfests and German Christmas-themed events, but the rest of what made the German-Americans, well, "German" has been set aside for a globalist, easily palatable culture.
