The way I'd see a Rider route playing out would focus on the man/monster/hero trichotomy from Fate/HA. Rider, who thinks of herself as a monster, would be squared off against Gil, the ultimate hero, and believe she has no chance of victory. Shirou, as man, either intervenes directly, defeating Gil but not saving Rider; convinces Rider to cast aside the trichotomy and face down Gil together as man and woman toppling the hero; or embrace the trichotomy himself and his transcendence of the human condition, and go down in a blaze of glory together with Rider as two monsters slain by the hero.

When you think about it, Shirou summoning Caster would have been an effectively unstoppable combination.
Shirou would win her over extremely easily with his housewife skills, and with Casters reinforcement magic and proper tutelage in using his Reality Marble would be an great front line fighter. She's by far the best informed of the Servants, knowing pretty much everything from the get-go. Shirou without her makes Gilgamesh look like a bitch, so their only problem is Berserker and maybe Lancer.
