I'm thinking that Choromatsu wrote that he wanted someone (Oso) to stop him.
From what it seems like how it's being set up, the letter will spur Osomatsu into action, mirroring EP 2 (technically the first) where he's looking his brothers, but this time trying to get them back together instead of looking for someone to play with. It seems like the most obvious route to go though, there are lot of ways the next episode can play out.
I'm simultaneously hoping and dreading that this might be the last episode we'll ever have. If they can maintain a certain level of quality, I'd be fine for the show going on forever like Gintama has, but it might be better for it to end properly. I'm looking forward to next week with a heavy heart.

Anyone who thinks this won't get a second season is delusional. We'll probably get closure to today's episode and an announcement at the end about the next season being planned. Even if Pierrot hates money and doesn't make another season there will be at least an OVA or two, possibly a movie. But with how popular the show is it'd be crazy to not make another season.
Hopefully it ends with Oso starting to get his act together and the brothers still getting together for shenanigans. At the minimum I hope Choro keeps his job- one of the brothers has to grow up.
