Do the NPCs that can become your masters just go away as you progress in the main storyline? I saw Piccolo for a bit but didn't want to be his master but now he's gone and Vegeta is available. Do I just miss out on getting his skill or something now?

Only one of them spawns somewhere in town at any given time. Which one it is resets every time you enter the character select for PQs or PvP.

They appear randomly. You can never permanently miss anyone but as more masters become available it's harder to get the specific one you want to spawn.
