When the blood curse was lifted in Revised, yes. Assamites were the fucking scum of the earth because there was no clan weakness other than "you really want to suck the blood/power out of every vampire." Basically a pass to be edgy as fuck.
Toreador was a problem too, though mostly from personal experience. Fat attention whores always wanted to play Toreador. I don't think a single fat chick I played with didn't choose Toreador, besides one, who chose Daughter of Cacophony (which is basically just a Toreador/Malk hybrid).

there's little things in there for every clan but I wouldn't say they're worth replaying the whole game for just for them (I replay the game a few times a year because it's my favourite). That said, Malkavian uplifts the whole dialogue, it's definitely worth it

Play The Final Nights as a Samedi if you want an actual Nossie experience. You HAVE to use the sewers or Obfuscate to get around. The NPC panic distance is much larger. There is a small grace period when changing areas though, so you won't get a masquerade violation just for stepping out of the diner.

If you feel like roleplaying Nos in your first playthrough, do it. It doesn't change the game quite as much as malk (which you should leave for second playthrough, always). Plus, it's not like you have to use the sewers, every game a Nos I just run in the streets. Unless you aggro 3 or more people in a short time span, you can't break the masquerade.
You also get a bunch of unique Nosferatu dialogue that you can tell is targeted towards you and no other clan. That made a tremendous first impression on me the first time I played the game, as it shows how deeply branched the dialogue system is.
