Highly doubtful, unless for whatever reason Sony decides to put him on stage, but Nier is way too niche for Sony's E3 press conference. Something like Persona 5 might get that air time, but Nier doesn't have a chance in hell.
Although he might show up in the Square-Enix Presents all day live stream things, but that's nothing like the absurdity of him showing up at a press conference.
