How varied are the characters bosses and storylines? I know there's some overlap but I'm hoping to see plenty of new bosses down the road

There are bosses unique to certain characters. Besides Velvet who you fought you'll also fight other characters along the way.

The bosses are mixed. Some are shared others are unique. I believe in boss rush mode you can tackle all the bosses as any character.
I haven't cleared Gwen's story yet myself, I think I'll start pacing myself at 1 character a day starting tomorrow, but in the original game all of the characters played fairly differently. Mercedes and Velvet were the most different, but each character has their own little gimmick that helps them get by. Not sure how it is in Leifthrasir but I imagine that with the new skills each character has a bit more flavor to them.

They changed the gameplay to be more like Muramasa (along with having the original game present). It's pretty much the same otherwise, I think.
