Did it get too big? Did it become too much of a convention instead of a charity event? Did too many speedrunners use GDQs as platforms to springboard themselves into twitch personalities?

When anything gets too popular it starts getting watered down and shitty. This is generally because organizers seek even greater popularity and the power/wealth that comes with it. They're also more likely to give in to what appears to be popular opinion, which is generally whining about inconsequential things that add up to be a culture. When these things are removed then the community appears sterile.

Speed running will always be a thing
always. its what people enjoy doing. difference is we'll more then likely only hear about big world record breaks or new games popping up that are pretty fun to speed run.
AGDQ is self is a joke, half the donations they "read" are made up on the spot or are completely fake. 2013 the same donation was read 6 times, I highly doubt the same person donated several times just to hear his name / message. as for the money going to charity I haven't been able to dig anything up. all I got is no one wants to talk about it.

The big personalities of the past like Cosmo and Siglemic are gone and have no replacements. GDQ has been watered down into a cold, mass market product that lacks the appeal of previous events.
