My runner-ups are personal choices. Superbad is the most objectively best movie from Seth Rogen and crew. I like Funny People because I like comedians and the movie is just them bullshitting. This is the End had me in stitches both times I saw it. Pineapple had some good laughs, but they weren't good enough at ad-libbing in it. It might be, since you seem to appreciate the comedy on a level I wasn't able to. That's not to say I didn't find Playtime funny or failed to understand any of the humor, I just enjoyed the visual design and rhythm of the comedy more than the actual humor itself, if that makes sense.
