I'm almost 100% positive that it's just to homage this comic cover from the DD comics. I think it's actually part of a larger story arc involving Punisher too, but don't quote me on that, I haven't read the comic in a while. Pic related obviously.
Also, hell yes, agreed that this fight blew the fuck out of the first season fight, and it's probably one of the best fight scenes I've seen in a while.

Basically, Daredevil knew there wasn't any bullet, that's why he laughed. He was laughing at the bikers for being intimidated by him, and probably getting a thrill out of what was about to happen. Because, ya know, Daredevil.
pretty sure he smiled because, well when you point a gun at someone, and pulll the trigger, and you have no bullets left you are basically telling the opposition to attack. you have no bullets, what have they to lose??
basically the only thing keeping those thugs from charging him was that gun, when he pulled the trigger (revealing to the thugs that he had no bullets) he was basically pressing the "start fight" button

I felt like that fight was embarrassingly obvious in it's attempt to recapture the magic of the season 1 fight, and it didn't even come close.
