I'm pretty sure I have all characters. The reaper is fairly well hidden. I'll spoiler it bit by bit in case you want to figure it out yourself at some point.
1.Just outside of Canterlot there's a hidden path to the left, leading to a big overworld maze which is a bit of a pain to get through. If you follow the maze to the end, you'll find a level called "The Hole".
2.If you go through the level normally, all that will happen is you'll fall down a hole, find a lot of extra lives and leave the level. But there's more to it.
3.When falling down the hole, there's a breakable wall to the left. Break it and go in.
4.Within the discovered path there is a door you can't see because the wall is blocking your view. Just press X at the end of the path and you should go in. The rest is self-explanatory. You'll find a chained up pony at the end, which is the character you unlock.
What's this message on the right? Maybe I can use it as a password. I can't into wingdings weblings dingalings.

Is Twilight in Alicorn Princess form supposed to be unlockable? She doesn't show up on the character selection screen - I have to redo the unlock process every time i want to play her, and she's completely overpowered, killing every enemy and boss (except for the last one) in one charged up attack. Not to mention how many jumps, projectiles and HP she has.

Pretty sure it's on purpose. If you're playing on Impossible Difficulty, you still get killed in one hit by most enemies, but in terms of damage, Twilicorn with Fausticorn as AI-ally take out every boss in seconds.
