Act 5 was the point that doomed Homestuck, basically because Hussie got too enamoured with the trolls and the fan reaction to it. He hit the mystical Sonic OC equation, then rather than let them be minor characters that do their bit and shuffle off of the main story they hung around forever. Then he tried to do the same thing again with another set of kids, only these ones were even less interesting. Then another set. Then another set, each one losing more of what had made the kids and trolls compelling in the first place. Then bringing back some of the original characters whose arcs had completed satisfactorily in order to recapture the initial buzz (failing because that buzz relied on the early context, now lost). Then he tried rebooting the troll dynamic with two new characters, who were, respectively, angry Karkat and plot-dumper (one of several plot-dumper characters, in fact, so even more of a failure).
Simultaneously getting infected with Tumblr brain diseases from his new fan base making him want to deliberately alienate the older Whistles/Humanimals/PS-era fans, losing interest in the story of Homestuck, depression, his personal life going off track and being a complete moron about the whole kickstarter debacle killed any momentum there had been, turning what had been a fast-paced constantly inventive story into a dirge about self-absorbed arseholes.

Lots of people cite this as the panel that got them hooked, which makes total sense because it established that Hussie was capable of writing in a literary register and therefore we could expect a lot of variety out of the story.
But it also hinted at the mythology underlying the main narrative. There were other Act 1-4 plot elements that Hussie never followed up on, too, like the Ultimate Riddle, which seems to be directly connected to this passage. Why wasn't that the focus of late Act 6 and Act 7? You introduce something that's literally called "The Ultimate Riddle," develop tons of themes that could easily be related throughout the story, and then drop the ball that hard at the end? When we learned about LE later on, it seemed to me that he was a continuation of this plot thread, but in the end he turned out to be a strong gay cloun alien that wanted to kill his sister, not a force of nature or manifestation of entropy or anything interesting.

Universes (Genesis Frogs) all die eventually. Reality (Paradox Space) "wants" to keep existing, and for that purpose universes all come with universe-creating magic that will let them reproduces (SBURB).
Paradox Space contains Horrorterrors and various iterations of Skaia. Whether they are brought there/created by the game, or natural inhabitants, I forgot. Maybe we don't know.
Skaia wants new universes, H.T.s don't. The game itself, with its players and bosses, is a bargain between the two parties: if the players win, Skaia wins, otherwise the Horrorterrors win.
I'm not sure how involved Skaia and the HTs are. Whether they're disputing a chess match and moving the pieces themselves, or stepping back to let the game take its course (like a particularly elaborate bet), I'm not sure.
Questions like "who made the game" only lead to either more questions in the vein of "who made God?" (like "where do those guys who made the game come from?") or causality loop bullshit. You don't need to know the exact origins of SBURB to enjoy the setting.

There are many answers to that question.
Sburb is based on data contained from the Frog Temple – which is itself a game construct, forming a causal loop.
It's also a wager between Skaia and the Horrorterrors, for the right to create a universe – but Skaia is also a game construct, which makes that another loop.
It's the natural reproductive cycle of a Genesis Frog – but they all exist within Sburb sessions, which makes that another loop, unless they existed in some other environment beforehand.
Ultimately, Sburb itself might have no "true" point of origin; it, and the universes that create it, may exist entirely as self-initiating time loops. Paradox Space is full of things like that.

As mentioned before, every session of the game is a wager between the forces of Creation (Skaia) and Destruction (the Horrorterrors) for the right to create a new universe, using a series of proxies such as the Armies of Light and Darkness, the Players, and the Denizens. The outer gods of the Furthest Ring are opposed to orderly existence and creation.
This is important to remember when asking why Sburb is the way it is. While it's never stated outright, it's likely that the extinction of a species is a price that Destruction demands of Creation as part of each session. It's worth noting that the Trolls's interstellar civilization could have survived the death of their host planet, were it not for the Vast Glub emitted by Gl'bgolyb, an "emissary to the Horrorterrors".
