If you could write about Karamatsu becoming uncharacteristic depressed, dropping the cool act and self depreciating. Then the brothers blaming Ichimatsu and guilt tripping him to make him bring Kara back to normal. Maybe with him starting with with half assed attempts off cheering Kara but keeping on failing and all escalating as he becomes guiltier and even trying to copy the usual Kara to make him happy again. All ending into a happy conclusion with some tsundere Ichi and dere Kara would be really nice.

I think this sort of show (gag anime, stylized art, etc) is simply better suited for fanart than for fanfiction. And if one does write fanfiction it's not quite that easy to balance a possibly more serious plot with the show's typical humor.
That said, I'm craving good fic like you wouldn't believe. If you're still taking requests, perhaps something taking place during high school? General shenanigans (with optional Ichi/Jyuushi?) would be nice!

are both alphas, Kara, Choro and Totty are all betas and Ichimatsu unfortunately for him, is an omega.

so he can let him know that Ichi is HIS omega and that as a beta Kara should step off.

Same appeal as homestuck and kancolle and 2hu. Large cast of samey characters each distinguishable by a simple gimmick. It's the ideal formula for making fanwork and shipping. Being primarily comedy based interspersed with genuine moments of emotional drama is also good shipping fuel. It tells the fans just enough about the characters' personalities and relationships with each other that they can fill in the rest themselves, like playing with dolls in a doll house.
