It's hard to explain, someone smarter than me could probably do a better job of it. There's something about the pacing and comedic timing afforded by the format (and Yotsuba's specific sort of comedy) that is really difficult to replicate in animation. Look at a lot of the criticisms of Nichijou for an example. I'm not saying it would be the exact same thing with a Yotsuba adaptation, but the parallels are there.

The story of Yotsuba is the story of a little kid and her friends and the jokes are realistic and grounded. It doesn't work that well in anime because the jokes take away from the format. An easy way to understand this is how fast you can read a chapter. Basically a couple of minutes. Any anime episode is essentially 20 minutes long. Not hard to see why it doesn't work.

Azuma didn't fully like the end result of how the azumanga daioh anime. He is even more possessive and protective of yotsubato. Unless he is fully confident in ending up perfectly to his vision he wont do it.
It is near impossible to perfectly translate a mango to an animu. So unless a production company is willing to put no budget or time limit constraints and is willing to hire the best in the industry there is no way a Yotsuba anime will be made.
