The charge used in the manga is likely pure bullshit for the time period and just used for dramatic effect but if it did exist it would be far too large for someone to throw and would probably be far larger than what is shown to do the damage it was supposed to.
A greek fire incendiary grenade is a simple clay or glass vessel filled with oil that has a wick. For an explosive to do damage it has to be pretty specially made. Especially if all you have access to is black powder.
But long story short the whole idea of it is not too far fetched but just enough to raise doubts.
