My guess is that enhancement is the ability to amplify the body's processes through some mumbo-jumbo like metabolism. To actually age would require a ludicrous amount of nen (and no enhancers we've seen in the series is confirmed to have the same potential as Gon's own, which could be rather accurately described as "bottomless"), and to channel that much nen would require a single mindedness that is really only seen in humans in the very stupid and those suffering from absolute despair.

From the very beginning of the manga (i.e. the first chapter), it was established that Gon was not a normal kid. He's impulsive and hardly thinks of the consequences of his actions. Gon-san was the epitome of that.
Many times during the story, other characters comment how uncanny his personality is, and the fact that nobody else tries what he did to defeat Pitou is just to emphasize this specific fact.
Of course, this is from a narrative point of view. The story is not finished yet, and there's nothing in the internal rules of the nen system that could prevent a second Gon-san.
