Ruroni Kenshin
Kenshin is like, 34 or 35. though he looks like he's still in his 20s (played for laughs in a couple of offhand lines) season 1 and 2 are pretty good, season 3 starts getting ridiculous pretty fast though...
it's a manga, but BERSERK features a heavily grizzled main character in what I'd assume is his mid to late 30s.
Ghost in the Shell. the major is in a young body, but she is far from young
granted, I'm not counting anime where characters are a supernaturally old, but justify their nature to act like children such as Hellsing or
Trigun
thats the best I got, what do you have on your list already?
I'm curious...
I'm pretty sure the guys in mouryou no hako are mid 30s

There are some anime adaptations of Berserk, though all of them put together don't come close to covering the entire story.
Manga would generally be a better place to look for older protags - for example, Jiro Taniguchi's works typically feature main characters somewhere in the 30-50s age range.

Gintama has quite a few older main characters. If you ignore Shinpachi and Kagura, the main cast has an average age of about 30.

rurouni kenshin has 28
This might blow your mind, but there's a ZoE anime and it stars a middle aged delivery man. It's pretty darned unusual to say the least.

Black lagoon is a good one. I'm pretty sure the ages range from 20s to 30s.
