If you want to improve the quality of your sprite, You need to look up pixel art and art tutorials, and practice.

That's a whole other kettle of fish, mate. I've got no experience with pixel art, but I think it would be best to get your artstyle down before worrying about animation.
For some good inspiration, you should check out some Way Forward games like the Shantae series and the switch force games. They have some great female walking and running animations.
