I'm pretty sure I killed Jessup's crew in a playthrough, and I was still able to do business with the Khans. But I might be misremembering.

I think you can overcome the rep loss just by helping out the regular Red Rock Khans. If you aren't planning on aiming for peaceful resolution, all you miss out on is an unarmed technique and a couple quests.

nope. you can go right on over to red rock and do the khan quests. only thing you need from him in information on benny then your free to do as you will with him. when you kill anyone in this game, just make sure you pump them for information first.

I really hope you've got 8 END for Implant GRX or you're missing out on the best drug in the game.

Had a lot of fun with hardcore mode, melee & unarmed only, survival to heal and no medicine or chem use. This was obvs more fitting for a Caesar run.
