You have to wait til lock on. You ult, wait for the red reticles. When they close, I think they even turn into skulls, you`ll get a kill on that target. Otherwise you`ll do damage based on how long the reticle was on them. That`s the catch of the ult, you announce it out loud then have to take a while before actually using it if you`re going for multiple targets.

How Deadeye works is that the damage on the shot ramps up every second you have a lock on (it's something like 180 HP every second). This means if they are squishy or are already weak it will be a kill shot quicker. This also means that sometimes its worth it to fire early because using Deadeye doesn't actually drain your Ammo, so you can just chunk them for a bunch of damage and finish them off with a Fan.
Also because there's limit to how high the damage can scale too, this means you can potentially one shot a Reinhardt shield and murder everyone behind it (got a pretty funny double kill that way once)
