The "This will be the last patch" was a mistranslated statement, but it was roughly meant to be "this is the last patch the game will get balance-wise."
And the way he talked about who was getting into the game pre-Mewtwo reveal basically said "There will be no other characters" just by way of him saying how he decides on who actually gets into the game.
In short, he's lied, but it doesn't seem as if he's ever lied to us if the intent wasn't to hype us the fuck up.

The way Sakurai sounded, I highly doubt that'll happen, just the way he rattled off all the stats on characters/stages/trophies/etc. at the end.

I'd rather they make Squirtle and Ivysaur fleshed out characters honestly, I wasn't a fan of Pokémon Trainer's "you have to keep changing forms or you'll get weaker and weaker" gimmick.
I'd personally like to see a Mortal Kombat Trilogy-style approach where the next game brings back all the characters and only adds a few newcomers compared to an entry that cuts characters and adds a ton of newcomers, just because we're starting to run out of potential first-party newcomers that Sakurai doesn't hate. Maybe redo the clones to be more like Roy in Smash 4 (clearly based on Marth but his properties are wildly different with some changed moves), that way Marth/Lucina/Roy and Link/Young Link/Toon Link wouldn't be too redundant.
