Fontaine had the workers unions (which technically were illegal in rapture), the mob and an entire underclass of trash willing to do his bidding. Ryan banning religion, as well as pretty much everything he didn't like, created a massively profitable black market for Fontaine to exploit. Then he got Tenenbaum on his payroll and created Jack.

Well the problem was there was an entire plot written for the game, but then Levine decided fairly last minute to fuck with it. Finktown I'm sure had a completely logical conclusion that led into the action of the Vox revolution, but instead they decided "WHOAH WHATS WITH THIS CUHRAZY TIMELINE STUFF?? WELL EVERYONE WANTS TO KILL YOU NOW, BETTER RUN!"

Finktown didn't even conclude, the story literally just changed to a different one 3/4 of the way through the level

as for actual medication for splicer therapy I think that's what Tenenbaum was trying to make in the first few games. There's a audiolog in 2 from her which says that "right now it can only remove the slugs from the little sisters" but she's trying to work on a tonic to combat splicer sickness
