Duke is an odd character, he's essentially a parody of 80s action stars. With that being said given the right care you could totally make a great Duke game as long as it's somewhat self-aware of how out of time Duke is. DNF tried to act like it was a come back from the good ol' days in reality it wasn't. It was like a depressing reunion tour of a classic band. Duke can be saved if given care. It was like that one kid who thought fart jokes were the epitome of humor, and you met him ten years later to realize that he was still making fart jokes.
You can go play Duke 3D today and realize that whoever wrote it had to be pretty clever to set up some of the jokes in that game specifically so the player would see them and get the punchline.

In regards to the OP: I wouldn't hold my breath. George Broussard mismanaged the project so hard the lost both the DNF license and the license to future Duke games.
Considering that Gearbox holds the license, I wouldn't expect them to pay attention to the Duke franchise until they run out of publishers to fleece or Borderlands stops laying golden eggs.
